Chestnut is an important woody food crop,Due to its high nutritional value and tolerance to poor soil,Widely planted in mountainous areas。Research on chestnut in recent years has mainly focused on trait correlation analysis、Molecular marker development and resistance to chestnut blight,Lack of genetic structure analysis and gene function verification for traits directly related to yield (seed size) and indirectly related traits (leaf length)。In recent years, our research group has published the first genome of chestnut,Constructed an instant transformation system for chestnut callus,Laying the foundation for rapid identification of gene functions。
151 Chinese chestnut varieties (lines) were extensively collected in this study,and perform simplified genome sequencing。Population genetics results show,Chinese chestnut varieties can be divided into two ecological types, north and south。Through bet365 online casinogenome-wide selection signal analysis of two ecotype populations,Identified tandemly arranged genes that may affect chestnut traits and environmental adaptability。To further explore the formation of chestnut yield-related traits,Conducted a genome-wide association study on 6 important horticultural traits in 151 varieties (lines)。45 highly related QTL identified,Sifted out 6 genes that are highly related to these traits,Including the candidate gene CmAP2 that affects chestnut grain weight and the candidate gene CmCIB1 that affects leaf length。Use gene silencing and overexpression technology to verify its function,Discovered that CmAP2 affects seed size by negatively regulating cell size,CmCIB1 affects the length of leaves and shoots by affecting the length of cells。
